[Main Ingredients] Neostigmine Methylsulfate
[Description] This drug is of colorless transparent liquid.
[Pharmacological Action] Neostigmine inhibits cholinesterase activity, preventing the hydrolysis of acetylcholine and increasing its concentration in the body, thereby enhancing and prolonging the effects of acetylcholine. It has strong excitatory effects on the smooth muscles of the gastrointestinal tract, bladder, and uterus, while its effects on glands, iris, bronchial smooth muscle, and cardiovascular inhibition are weaker. Its central nervous system effects are not significant.
Neostigmine has the strongest effect on skeletal muscles. In addition to its cholinesterase-inhibiting action, it directly excites N-receptors at motor endplates and promotes the release of acetylcholine from motor nerve terminals.
Oral administration of neostigmine is poorly absorbed and does not easily cross the blood-brain barrier. Plasma protein binding rate is 15% to 25%. Some of the drug is hydrolyzed by plasma cholinesterase and excreted via urine as quaternary ammonium alcohol and unchanged form. Metabolized portions are excreted via the bile.
[Drug Interaction] (1) This drug can prolong and enhance the muscle-relaxant effects of depolarizing neuromuscular blockers such as succinylcholine chloride.
(2) It exhibits antagonistic effects with non-depolarizing neuromuscular blockers.
[Function and Use] Anticholinesterase drug, primarily for the treatment of gastrointestinal hypotonia, myasthenia gravis, retained placenta, etc.
[Dosage and Administration] Intramuscular and subcutaneous injection: 4-10 ml/time for horses; 4-20 ml/time for cattle; 2-5 ml/time for sheep and pigs; 0.25-1 ml/time for dogs.
[Adverse Reaction] Side effects are minimal at therapeutic doses. Overdose may cause sweating, bradycardia, muscle tremors, or muscle paralysis.
[Precautions] (1) It is prohibited for animals with mechanical intestinal obstruction or bronchial asthma.
(2) Atropine can be used to counteract the excitatory effects on M-receptors in cases of poisoning.
(3) This drug can prolong and enhance the muscle-relaxant effects of depolarizing neuromuscular blockers such as succinylcholine chloride; it exhibits antagonistic effects with non-depolarizing neuromuscular blockers.
[Withdrawal Period] It is not required.
